RESUMO

Aim: 7-Ethyl-10-hydroxycamptothecin (SN-38)-loaded gold nanoshells nanoparticles (HSP@Au NPs) were developed for combined chemo-photothermal therapy to treat colorectal cancer. Materials & methods: SN-38-loaded nanoparticles (HSP NPs) were prepared by the lyophilization-hydration method, and then developed into gold nanoshells. The nanoparticles were characterized and assessed for photothermal properties, cytotoxicity and hemocompatibility in vitro. In vivo anticancer activity was tested in a tumor mouse model. Results: The HSP@Au NPs (diameter 186.9 nm, zeta potential 33.4 mV) led to significant cytotoxicity in cancer cells exposed to a near-infrared laser. Moreover, the HSP@Au NP-mediated chemo-photothermal therapy displayed significant tumor growth suppression and disappearance (25% of tumor clearance rate) without adverse side effects in vivo. Conclusion: HSP@Au NPs may be promising in the treatment of colorectal cancer in the future.

RESUMO

In this study, ε-polylysine and calcium phosphate precipitation (CPP) methods were employed to induce antibacterial effects and dentin tubule occlusion. Antibacterial effects of ε-polylysine were evaluated with broth dilution assay against P. gingivalis. CPP solution from MCPM, DCPD, and TTCP was prepared. Four concentrations of ε-polylysine(ε-PL) solutions (0.125%, 0.25%, 0.5%, 1%) were prepared. Dentin discs were prepared from recently extracted human third molars. Dentin discs were incubated with P. gingivalis (ATCC 33277) bacterial suspension (ca. 105 bacteria) containing Brain Heart Infusion medium supplemented with 0.1 g/mL Vitamin K, 0.5 mg/mL hemin, 0.4 g/mL L-cysteine in anaerobic jars (37 °C) for 7 days to allow for biofilm formation. P. g-infected dentin specimens were randomly divided into four groups: CPP + 0.125% ε-PL, CPP + 0.25% ε-PL, CPP + 0.5% ε-PL, CPP + 1% ε-PL. On each dentin specimen, CPP solution was applied followed by polylysine solution with microbrush and immersed in artificial saliva. Precipitate formation, antibacterial effects, and occlusion of dentinal tubules were characterized in vitro over up to 72 h using scanning electron microscopy. ε-PL showed 34.97% to 61.19% growth inhibition levels against P. gingivalis (P. g) after 24 h of incubation. On P. g-infected dentin specimens, DCPD + 0.25% ε-PL, and DCPD + 0.5% ε-PL groups showed complete bacterial inhibition and 78.6% and 98.1% dentin tubule occlusion, respectively (p < 0.001). The longitudinal analysis on fractured dentin samples in DCPD and TTCP groups revealed deeply penetrated hydroxyapatite-like crystal formations in dentinal tubules after 72 h of incubation in artificial saliva.

RESUMO

The causes of high biological diversity in biodiversity hotspots have long been a major subject of study in conservation biology. To investigate this matter, we conducted a phylogeographic study of five Drosophila (Diptera: Drosophilidae) species from East and Southeast Asia: Drosophila albomicans Duda, D. formosana Duda, D. immigrans Sturtevant, D. melanogaster Meigen, and D. simulans Sturtevant. We collected 185 samples from 28 localities in eight countries. From each collected individual, we sequenced the autosomal extra sex comb gene (esc) and seven mitochondrial genes, including nicotinamide adenine dinucleotide hydrate-reductase dehydrogenase subunit 4 (ND4), ND4L, tRNA-His, tRNA-Pro, tRNA-Thr, partial ND5, and partial ND6. Phylogenetic analyses using maximum- likelihood and Bayesian methods revealed interesting population structure and identified the existence of two distinct D. formosana lineages (Southeast Asian and Taiwanese populations). Genetic differentiation among groups of D. immigrans suggests the possibility of endemic speciation in Taiwan. In contrast, D. melanogaster remained one extensively large population throughout East and Southeast Asia, including nearby islets. A molecular clock was used to estimate divergence times, which were compared with past geographical events to infer evolutionary scenarios. Our findings suggest that interglacial periods may have caused population isolation, thus enhancing population differentiation more strongly for some of the Drosophila species. The population structure of each Drosophila species in East and Southeast Asia has been influenced by past geographic events.

RESUMO

Recent advances in material science and technology yield not only various kinds of nano- and sub-micro-scaled particles but also particles of various charged conditions such as Janus particles. The characterization of these particles can be challenging because conventional electrophoresis theory is usually based on drastic assumptions that are unable to realistically describe the actual situation. In this study, the influence of the nonuniform charged conditions on the surface of a particle at an arbitrary level of surface potential and double layer thickness on its electrophoretic behavior is investigated for the first time in the literature taking account of the effect of double-layer polarization. Several important results are observed. For instance, for the same averaged surface potential, the mobility of a nonuniformly charged particle is generally smaller than that of a uniformly charged particle, and the difference between the two depends upon the thickness of double layer. This implies that using the conventional electrophoresis theory may result in appreciable deviation, which can be on the order of ca. 20%. In addition, the nonuniform surface charge can yield double vortex in the vicinity of a particle by breaking the symmetric of the flow field, which has potential applications in mixing and/or regulating the medium confined in a submicrometer-sized space, where conventional mixing devices are inapplicable.

RESUMO

The high accuracy of multidetector-row computed tomography (MDCT) in evaluating prosthetic valve disorders has been confirmed. This, we believe, is the 1st report of the use of MDCT to detect and evaluate left ventricular outflow tract (LVOT) pseudoaneurysms in patients who have undergone aortic valve replacement with prosthetic valves. We used MDCT to scan 21 such patients, 3 of whom had a small pseudoaneurysm in the LVOT. Each pseudoaneurysm projected away from the LVOT and had a narrow neck that was located just below the sewing ring of the prosthetic aortic valve. One pseudoaneurysm was not thrombotic, 1 was partially thrombotic, and 1 was completely thrombotic. One of these had gone undetected earlier on transthoracic echocardiography. We consider MDCT to be superior to echocardiography in the detection of LVOT pseudoaneurysms in patients who have undergone aortic valve replacement with prosthetic valves. We publicize our results in the hope that larger studies will be undertaken in order to investigate the prevalence and clinical implications of our findings.
